Chlamydia trachomatis is an important cause of sexually transmitted infection that can manifest itself as either acute cervicitis, pelvic inflammatory disease, and more commonly as a chronic asymptomatic infection. Previously we reported Ag-PVP (5-10 nm) reduced major pro-inflammatory cytokines produced during C. trachomatis infection of mouse J774 macrophages.Here we tested the role of Ag-PVP on the expression level of Toll like receptor (TLR) and NOD like receptors. We employed qRT-PCR to determine the expression level of TLR2 and NOD2 using RNA extracted from C. trachomatis infected mouse macrophage cell pellets that were exposed to Ag-PVP (2.5 µg/mL). Our result shows that Ag-PVP down-regulated the expression of TLR2 and NOD2, suggesting that Ag-PVP may exert its anti-inflammatory properties by interfering with TLR2 and NOD2 expression. We next investigated the anti-inflammatory role of Ag-PVP on primary cells employing bone marrow-derived macrophages (BMDM). Ag-PVP was relatively non-toxic to BMDM (up to 6.25 µg/mL) over a 72-hour time-period. Further studies revealed, Ag-PVP (2.5 µg/mL) inhibited the production of IL-6 and CXCL10 by C. trachomatis-infected BMDM as determined by ELISA. Overall our data provides further evidence for Ag-PVP role in reducing cell surface receptors expression to regulate C. trachomatis inflammation in macrophages.
