The different biological activity between the colloidal silver nanoparticle and the plasma silver nanoparticle in various cell types

Abstract:Purpose: The objective of this study was to exam any difference after treatment of nanoparticle in cellular proliferation and gene expression in various cell types. Particularly, it was compared between the colloidal silver nanoparticle and the plasma silver nanoparticle. Materials and Method: SCC-9 (oral cancer cell line), I2 (chondrocyte), and primary cultured periodontal ligament cells were used in this study. The concentration between 1 g/ml and 20 g/ml was used. The cellular proliferation was evaluated by MTT assay. The gene expression after treatment of silver nanoparticle was studied in I2 cells by cDNA microarray. Results: Regardless the type of silver nanoparticle, the application of silver nanoparticle showed the decreased value in MTT assay. Generally, the effect on cellular proliferation was prominent in the colloidal silver. Interestingly, SCC-9 showed more resistance in all used concentration in both types of silver nanoparticles. The gene expression showed different profile between the colloidal silver nanoparticle and the plasma silver nanoparticle. Conclusion: Though the same material was used for generation of nanoparticle, the method could influence their biological activity. The biological activity of silver nanoparticle was also different in the different kind of cell type.
